Abstract

Fluorochromes in combination with flow cytometry can be used for laboratory assessment of semen quality in humans and domestic animals. Some studies have reported the potential toxicity of these fluorochromes toward the cells analyzed, but not toward the laboratory technician who operates the analytical instrument. We tested the genotoxic potential of three fluorochromes, SYBR-14, propidium iodide, and Hoechst 33342, using the colorimetric SOS ChromoTest?. The test revealed no genotoxic potential for any of the three fluorochromes within the dilution ranges investigated. We conclude that occasional direct contact with these fluorescent probes does not necessarily pose a genotoxic hazard.  相似文献

The problem of cryptic species in Diacyclops bicuspidatus was examined using interpopulation crosses of four populations collected from a: (1) permanent flood lake in Kiev, Ukraine, (2) temporary pool in Kiev, (3) permanent pond in St. Petersburg, Russia (1200 km to north from Kiev) and (4) lake in Crimea (1100 km south of Kiev). The only interpopulation crosses to exhibit fertility were those between the St. Petersburg population and each of the two Kiev populations. The crosses between the Kiev and Crimea populations, between the St. Petersburg and Crimea populations, and between the two Kiev populations were sterile, as evidenced by either nonviable eggs, empty egg membranes or incomplete copulations. The F1 hybrids resulting from the St. Petersburg permanent pond X Kiev flood lake cross were fertile and produced mature F2 offspring. Some data on development times of parental and hybrid lines are presented. The St. Petersburg parental line showed development times almost twice as long as those of the Kiev flood lake population when reared at 10 °C and 20 °C in the laboratory. The F1 offspring of the cross between St. Petersburg females and Kiev floodlake males showed similar development times to females of the St. Petersburg parental lines at both temperatures. The F2 hybrids also showed development times that approximated those of the St. Petersburg parental line. These crossbreeding studies suggest the presence of cryptic species in the D. bicuspidatus inhabiting ecologically different populations in many parts of its large holarctic range.  相似文献

The birth rate of natural cladoceran populations can change rapidly (during 2–3 days), reflecting rapid changes in their environment. If the egg ratio is calculated on the basis of egg age distribution, the birth rate can be estimated at short sampling intervals (shorter than egg stage duration) by modified Paloheimo's (1974) formula. When female size structure and age of eggs in clutches at the beginning and the end of a sampling interval are known, death rates of ovigerous females and eggs in separate size classes can be determined and incorporated in birth rate estimates. All these methods have been employed using the data on the population of Diaphanosoma brahyurum from the lake Obsterno (North-Western Belarus) in July–August, 1992. The birth rate values computed by the proposed methods and Poloheimo's formula differed significantly in many cases. The accuracy of birth rate estimations from various calculation methods was tested using a computer simulation. The model contains the essential features of cladoceran life history: distinct egg, juvenile and adult stages, development of eggs and reproduction. The population was divided into 25 age classes, each of 1 day duration. Durations of the egg, juvenile and adult stages were set at 3, 6 and 20 days, respectively. The embryogenesis was divided into three egg stages, each of 1 day duration. Survivorship was set from 0.2 up to 1.0 for each age class. The survivorship and brood size were changed through each of five time intervals (days) that allowed to simulate an increase or reduction of population density. Fecundity, survivorship and egg stage duration remained constant during each of 5 days that assumed stability of an environment (this does not occur in nature). Nevertheless, the egg ratio, proportion of juveniles and birth rates were variable even under these circumstances. Computer simulations showed that Poloheimo's formula evaluates birth rate with the relative error of 62% and usually overestimates its values. We propose methods to decrease errors of birth rate estimations by 3.5–5.5 times.  相似文献

Three new bisbenzimidazole (BBI) compounds, which differ from Hoechst 33258 mainly by substitution of a N-dimethylaminopro-pylcarboxamide group in place of the N-methyl-piperazine ring, were studied for their DNA- and AT-base pair specificity as well as for their ability to be quenched by incorporated 5-bromodeoxy-uridine (BrdU). Each of them had DNA binding specificity comparable to or greater than that of Hoechst 33258 and each had a greater specificity for AT-rich regions than did Hoechst 33258. The dependence of fluorescence of new dyes on the BrdU-incorporation into DNA is different from that of Hoechst 33258 and related compounds with piperazine ring. The quenching effect is much weaker, and two of the new compounds (BBI-1 and BBI-2) even show somewhat enhanced binding (fluorescence) at lower concentrations. Certain BBI dyes without piperazine ring may have some advantage over Hoechst for accurate DNA [AT-specific] measurements. The piperazine ring appears to play an important role in the yet unknown mechanism of Hoechst quenching by incorporated BrdU.  相似文献

Bivariate flow karyotypes of chromosomes from sheep, cattle and pig lymphocytes and from a cattle-mouse somatic cell hybrid line were obtained using a dual laser fluorescence-activated cell sorter (FACS). Pig chromosomes were resolved into 19-20 peaks, indicating that most, if not all, pig chromosomes could be separated by this technique. Sheep chromosomes showed incomplete separation but three clear peaks, presumably representing the three large metacentric chromosomes, plus five other clusters were obtained. Cattle chromosomes showed poor separation but about four peaks could be distinguished, indicating that certain chromosomes could be sorted in this species. The use of cattle-mouse hybrids may enable other individual cattle chromosomes to be obtained. It is concluded that FACS separation will be a useful additional tool for gene mapping.  相似文献

Extracellular, single unit recording techniques were used to measure the responses of posterior lateral line nerve fibers to a 50-Hz dipole source that slowly changed its location along the length of the fish. The flow-field equations for a dipole source were used to model the pressure gradient pattern and thus, the expected excitation pattern along a linear array of lateral line receptor organs for different source locations. Finally, excitation patterns were similarly modeled along the left and right side of the fish's head for actual steps taken by sculpin in approach pathways to the 50-Hz dipole source. Spatial histograms of posterior lateral line nerve fiber responses to different locations of the dipole source could be predicted from pressure gradient patterns modeled from the flow-field equations, confirming that the modeling approach applied to behavioral results was a good predictor of excitation patterns likely to be encoded by the lateral line periphery. An examination of how modeled excitation patterns changed from one position to the next in typical approach pathways and how patterns differed between positions from which successful and unsuccessful strikes were launched suggests that approach and strike strategies can indeed be explained by the information available in excitation patterns. In particular, changes in the spatial distribution of pressure gradient directions (polarities), available only when the source is lateral (as opposed to directly in front of the fish), appear to enhance the ability of sculpin to determine source distance. The present work focuses on the role of pili present at the cell surface of Lactococcus lactis in bacterial adhesion to abiotic (hydrophobic polystyrene) and biotic (mucin-coated polystyrene) surfaces. Native pili-displaying strains and isogenic derivatives in which pilins or sortase C structural genes had been modified were used. Surface physico-chemistry, morphology and shear-flow-induced detachment of lactococcal cells were evaluated. The involvement of pili in L. lactis adhesion was clearly demonstrated, irrespective of the surface characteristics (hydrophobic/hydrophilic, presence or not of specific binding sites). The accessory pilin, PilC, and the backbone pilin, PilB, were revealed to play a major role in adhesion, provided that the PilB was present in its polymerized form. Within the population fraction that remained attached to the surface under increasing shear flow, different association behaviors were observed, showing that pili could serve as anchoring sites thus hampering the effect of shear flow on cell orientation and detachment.  相似文献

This study was designed to understand better if and how juvenile sporophytes of Macrocystis pyrifera can photoacclimate to high-light conditions when transplanted from 10 to 3 meters over 7 d. Acclimation of adult sporophytes to light regimes in the bathymetric gradient has been extensively documented. It primarily depends on photoacclimation and translocation of resources among blades. Among other physiological differences, juvenile sporophytes of M. pyrifera lack the structural complexity shown by adults. As such, juveniles may primarily depend on their photoacclimation capacities to maintain productivity and even avoid mortality under changing light regimes. However, little is known about how these mechanisms operate in young individuals. The capacity of sporophytes to photoacclimate was assessed by examining changes in their photosynthetic performance, pigment content, and bio-optical properties of the blade. Sporophytes nutritional status and oxidative damage were also determined. Results showed that juvenile sporophytes transplanted to shallow water were able to regulate light harvesting by reducing pigment concentration, and thus, absorptance and photosynthetic efficiency. Also, shallow-water sporophytes notably enhanced the dissipation of light energy as heat (NPQ) as a photoprotective mechanism. Generally, these adjustments allowed sporophytes to manage the absorption and utilization of light energy, hence reducing the potential for photo-oxidative damage. Furthermore, no substantial changes were found in the internal reserves (i.e., soluble carbohydrates and nitrogen) of these sporophytes. To our knowledge, these results are the first to provide robust evidence of photoprotective and photoacclimation strategies in juveniles of M. pyrifera, allowing them to restrict or avoid photodamage during shallow-water cultivation.  相似文献

Recent studies show that both marine and limnic microalgal species often consist of several genetically distinct populations. This is also valid for the nuisance freshwater algae Gonyostomum semen, which originates from acidic, brown water swamp lakes, but can nowadays also be found in clearer lakes with close to neutral pH. We hypothesized that the observed genetic differentiation among G. semen lake populations, reported in earlier studies, is connected to adaptation to local environmental conditions. In the present study we performed controlled laboratory experiments to test whether 12 strains originating from five lakes varied in their response to five to six different pHs, light intensities and DOC concentrations. Overall, growth (0.01–0.37 day−1) was observed over a wide range of light intensities and pHs, demonstrating high potential for photoacclimation and extensive plasticity of individual strains. Moreover, we found similar growth rates and consistent growth optima for specific pHs by strains from the same lake, suggesting genetic differentiation of populations into distinct phenotypes. However, observed strain specific preferences did not always reflect environmental conditions in the lakes of origin and provided limited evidence for the hypothesized local adaptation. Instead, the observed phenotypic differentiation may indicate resilient effects of founder events. We suggest that the wide phenotypic plasticity in this species enables it to thrive in fluctuating and variable environments, and may play a role in its ability to colonize new habitats.  相似文献
